شناسایی دو آلل BF2*13 و BF2*21 کمپلکس اصلی سازگاری بافتی توسط روش PCR در دو توده نژادی خزک و دشتیاری

چکیده

این پژوهش به منظور شناسایی دو آلل BF2*13 و BF2*21 ژن BF2 از کلاس I کمپلکس اصلی سازگار بافتیMHC  در دو توده نژادی استان سیستان و بلوچستان صورت گرفت. کمپلکس اصلی سازگار بافتی مجموعه ای ژنی می باشد که نقش تنظیمی را در سیستم ایمنی ایفا می کند. آزمایشات بسیاری ارتباط این مجموعه ژنی را با مقاومت و حساسیت در مقابل انواع بیماری ها و همچنین صفات تولیدی نشان داده است. ژن BF2 یکی از این جایگاه های مرتبط می باشد. در این تحقیق، نمونه های خون بطور تصادفی از 72 قطعه مرغ بومی (37 قطعه مرغ خزک و 35 قطعه مرغ دشتیاری) گرفته شد. بعد از استخراج  DNA از نمونه های خون با کمک کیت استاندارد، کمیت و کیفیت  DNA توسط ژل آگاروز 1% تعیین گردید. مرحله بعدی، طراحی پرایمرهای مناسب برای آلل های BF2*13 و BF2*21 بود. با استفاده از PCR این آلل ها تکثیر شده و محصولات PCR توسط الکتروفورز روی ژل به ترتیب 52 و 197 جفت باز رویت گردید.   فراوانی نمونه های دارای آلل BF2*13 در دو توده نژادی خزک و دشتیاری به ترتیب 6/75% و 8/22% به دست آمد. همچنین فراوانی نمونه های دارای آلل BF2*21 در دو توده نژادی خزک و دشتیاری به ترتیب 9/45 و 2/34% مشاهده شد. از طرف دیگر، در توده نژادی خزک به رغم بالا بودن فراوانی نمونه های دارای آلل BF2*13، فراوانی نمونه های دارای آلل BF2*21 که رابطه مستقیم با مقاومت به بیماری های مارک، آنفولانزای مرغی، بورس عفونی و برخی صفات تولیدی دارد نیز در حد نسبتاً خوبی بود. بنابراین اصلاحگران می توانند این آلل ها را در برنامه های اصلاح نژاد مورد توجه قرار دهند.
