بررسی چندشکلی ژن‌های بتالاکتوگلوبولین، پرولاکتین و DGAT1 در گاومیش خوزستان

چکیده

تولید و ترکیبات شیر جزء دسته صفات کمی و چندژنی می‌باشند وتحت تأثیر تعداد زیادی ژن قرار دارند. این تحقیق به منظور تعیین چندشکلی ‌ژن‌های بتالاکتوگلوبولین، پرولاکتین و DGAT1، به عنوان ژن‌های کاندید مؤثر برصفات تولید شیر و ترکیبات آن،با استفاده از تکنیک PCR-RFLP درگاومیش‌های استان خوزستان صورت گرفت. از تعداد 200 رأس از گاومیش‌های استان خوزستان نمونه خون تهیه و استخراج DNA از نمونه‌ها به روش بهینه شده و تغییریافته استخراج نمکی صورت گرفت. بعد از انجام مراحل استخراج DNA، ژنوتیپ حیوانات با استفاده از روش PCR-RFLP تعیین گردید. برای تعیین ژنوتیپ حیوانات، قطعه 262 جفت باز از ناحیه اگزون و اینترون 4 ژن بتالاکتوگلوبولین، قطعه 411 جفت باز از اگزون 8 ژن DGAT1 و قطعه 156 جفت بازی از ناحیه اگزون 3 ژن پرولاکتین تکثیر و به ترتیب با استفاده از آنزیم‌های محدودالاثر HeaIII، CfrI و RsaI مورد هضم قرار گرفتند. محصولات هضم شده با استفاده از آنزیم، جهت تعیین ژنوتیپ بر روی آگارز 2% الکتروفورز گردیده و با اتیدیوم برماید رنگ‌آمیزی شدند. نتایج این تحقیق نشان داد که در گاومیش‌های مورد مطالعه آلل B در جایگاه بتالاکتوگلوبولین، آلل A در جایگاه پرولاکتین و آلل K در جایگاه DGAT1 تثبیت شده است و برای هیچ یک از جایگاه‌ها چندشکلی مشاهده نشد
