بررسی شبکه تنظیمی مؤثر در شروع شیردهی در بافت پستان گاو شیری با استفاده از داده‌های RNA-seq

چکیده

در این مطالعه به منظور درک بهتر شبکه تنظیمی درگیر درشروع شیر‌دهی در بافت پستان گاو شیری، در ابتدا ژن های با بیان بالا با استفاده از تجزیه و تحلیل داده‌های RNA-Seq در سه حالت زیر شناسایی گردید: حالت اول) مقایسه بافت پستان گاو شیری در 35 روز قبل از زایمان با 3 روز بعد از زایمان ، حالت دوم) مقایسه بافت پستان گاو شیری در 7 روز قبل از زایمان با 3 روز بعد از زایمان، حالت سوم) مقایسه بافت پستان گاو شیری در 7 و 35 روز قبل از زایمان. به منظور بررسی گروه‌های کارکردی ژن-های معنادار با بیان بالا در سه حالت مختلف ذکر شده، از نرم-افزار DAVID استفاده شد. تنها عبارات معنادار(05/0 >P ) مربوط به مراحل بیولوژیکی در نظر گرفته شد. سپس توالی پروموتری این ژن‌ها جهت شناسایی فاکتور‌های رونویسی کاندید درگیر در این مکانسیم استخراج شد. نتایج تجزیه و تحلیل پروموتری با استفاده از نرم‌افزار Genomatix منجر به شناسایی 24 فاکتور رونویسی کاندید جدید احتمالی مؤثر در مراحل تنظیمی شروع شیر‌دهی گردید. افزون بر این، برای ترسیم شبکه‌های ژنی مرتبط با فاکتور‌های رونویسی شناسایی شده در شروع شیر‌دهی از نرم افزار STRING استفاده شد. با توجه نتایج بدست آمده از این پژوهش، 24 فاکتور رونویسی جدید شناسایی شدند که دارای پتانسیل مناسبی برای بررسی بیشتر در سطح آزمایشگاهی می-باشند و می‌توانند اطلاعات جدیدی را در درک بهتر شبکه‌ی تنظیمی مؤثر در شروع شیر‌دهی در بافت پستان گاو شیری فراهم کنند.
