بررسی آزمایشگاهی اثرات وابسته به دوز آنتی‌بیوتیک سالینومایسین بر تخمیر شکمبه و برخی از جمعیت‌های میکروبی شکمبه با استفاده از روش Real-time PCR

چکیده

تحقیق حاضر به‌منظور بررسی آزمایشگاهی اثرات آنتی‌بیوتیک سالینومایسین در دوزهای مختلف (صفر، 2، 4، 8 و 16 میلی‌گرم بر لیتر) بر کینتیک تولید گاز، برخی شاخص‌های هضم و تخمیر شکمبه و در دوزهای صفر، 4 و 16 میلی‌گرم بر لیتر بر برخی جمعیت‌های میکروبی شکمبه با استفاده از روش Real-time PCR انجام شد. برای این منظور از سه رأس گوسفند نر فیستولادار جهت تهیه مایع شکمبه برای انکوباسیونهای 144 ساعته در مرحله اول و انکوباسیونهای 24 ساعته در مرحله دوم آزمایش استفاده شد. نتایج نشان داد که با افزایش دوز سالینومایسین گاز تولیدی در تمامی ساعات و حداکثر گاز تولیدی (A) بصورت خطی و درجه دو افزایش یافت. با این حال، نرخ تولید گاز ( µ) تحت تأثیر قرار نگرفت. همچنین میزان گاز تولیدی بعد از 24 ساعت (GP24)، میزان هضم آزمایشگاهی ماده خشک (IVTDMD) و ماده آلی (IVTOMD) و غلظت کل اسیدهای چرب فرار (TVFA) بموازات افزایش دوز سالینومایسین بطور خطی و درجه دو افزایش یافتند، اما فاکتور تفکیک (PF) کاهش یافت و توده میکروبی (MB) و غلظت آمونیاک تحت تأثیر سالینومایسین قرار نگرفتند. در بین میکروارگانیسم‌های شکمبه، جمعیت نسبی پروتوزوآها و فیبروباکترسوکسینوژنز در بالاترین دوز سالینومایسین کاهش یافت و جمعیت قارچها، متانوژنها و باکتری کلستریدیوم آمینوفیلوم تحت تأثیر سالینومایسین قرار نگرفت. این نتایج نشان داد که سالینومایسین اثرات مثبتی بر شاخص‌های هضم و تخمیر شکمبه‌ای دارد که با توجه به یافته های نوین مبنی بر اثرات آن در پیشگیری از انواع سرطان در انسان می‌تواند در تغذیه دام به‌عنوان افزودنی استفاده گردد.
