بررسی SNPهای مرتبط با چربی لاشه در گوسفندان نژادهای افشاری، مغانی و قزل

چکیده

شناسایی مناطق ژنومی کنترل کننده صفات اقتصادی، یکی از چالش برانگیزترین موارد استفاده از نشانگرهای متراکم در ژنتیک حیوانی است. شناسایی جایگاه‌های ژنی مرتبط با ذخیره چربی از لحاظ اقتصادی از اهمیت زیادی در پرورش گوسفند برخوردار است. در این تحقیق، از تعداد 49034 نشانگر تک نوکلئوتیدی برای شناسایی نواحی ژنومی موثر بر ذخیره چربی برای 106 رأس گوسفند از نژادهای افشاری (37)، مغانی (34) و قزل (35) استفاده شد. نمونه‌ها با استفاده از آرایه‌های Illumina OvineSNP50K Beadchip تعیین ژنوتیپ شدند. با بررسی کنترل کیفیت نمونه‌های تعیین ژنوتیپ شده از 49034 نشانگر SNP تعداد 46676 باقی ماند. برای قرار گرفتن حیوانات در گروه-های نژادی خود از آنالیز مولفه‌های اصلی (PCA) براساس اطلاعات روابط خویشاوندی ژنومی استفاده شد. جهت بررسی تمایز جمعیتی در بین این سه نژاد از آماره‌های تتا و FST استفاده شد. در این مطالعه پنج ناحیه ژنومی که در صدک 99/99 کل ارزش‌های FST قرار گرفته بودند برای بررسی‌های بیشتر انتخاب شدند. این مناطق بر روی کروموزوم‌های 13، 3 (دوناحیه)، 15 و 22 واقع شده‌اند که با صفات فعالیت گیرنده لیپوپروتئین با چگالی پایین، اتصال اجزاء لیپوپروتئین با چگالی پایین، تنظیم لیپولیز در سلول‌های چربی و متابولیسم اسید چرب مرتبط می‌باشند. برای ارزیابی نشانه‌های انتخاب بر پایه روش‌های عدم تعادل لینکاژی از آزمون هموزیگوسیتی هاپلوئیدی بسط داده شده استفاده شد. استفاده از تراشه‌های ژنومی برای مطالعه تفاوت‌های ژنتیکی گوسفندان بومی و بررسی صفات اقتصادی مفید بوده که این امر می‌تواند به دست‌یابی برای بهبود ژنتیکی از طریق مطالعات همبستگی ژنوم و انتخاب ژنومی کمک کند.
